We are wanting to explore the ability of human breast cell lines to exhibit circadian rhythm.  We are also interested in influence of environmental toxicants on circadian rhythm as well as general gene regulation.  I felt that the Kronos dio would be the proper equipment to perform this task as it has a CO2 hook up and can monitor 3 colors at once.  Therefore, I could transfect the BASIC luciferase construct using one color and the gene of interest using a different luciferase and monitor reporter activity in real time.

Principle of protein synthesis and purification

ExiProgen Protein Synthesis Kit useds a E. coli extract to effect coupled transcription/translation of input DNA, which can be plasmid, or PCR generated DNA. The protein itself is generated with a His-Tag, which is then purified using the Ni-NTA magnetic bead provided. The result is high yields of protein that is >90% pure.

Nucleic acid extraction principle

ExiProgen DNA/ RNA Kits work on the principle of cell lysis, followed by bind, wash elute from silica magnetic beads. High yields of ultrapure DNA or RNA are obtained with OD260 readings of > 1.8 for DNA and 2.0 for RNA.

Models

Heated Lid
The heated lid has a flush fitting enclosure to ensure a complete seal eliminating airflow across the sample block surface. The in-lid surface is spring-mounted to ensure even contact and pressure on all vials regardless of the tube height, ensuring no condensation of water vapor on the tube lids. A specially designed, microprocessor controlled heating element compensates temperature fluctuations along the lid.

Software
The control software installed in the T-CY has been designed to allow easy control and programming of the instrument. The control panel comprises a clearly visualized 168 character LCD, upon which navigation through the program menus is simple due to the logical file layering.

The layering of menus allows the operator to quickly select, view and start pre-entered programs. Each program can be displayed in full so that all parameters can be verified before commencing a program run.

The three elements required for a good thermal cycler are reliable performance, a fast uniform block and user friendly software. The MultiGene II combines a versatile and precise cycling unit with easy to use software to make it an outstanding personal thermal cycler. Not limited to the amplification of nucleic acids, the unit is also useful for applications such as enzymatic digestion, ligation, and other procedures that require incubation of small samples in a temperature controlled environment. 
Combining simplicity with performance, the MultiGene II features straightfoward and familiar programming with proven cycling technology in a modern molded housing. The compact design of the unit conserves bench space and allows it to be easily moved to different locations, such as under a biological hood, or stored out of the way when not being used.
Small enough to be considered a personal thermal cycler, the MultiGene II is supplied with either a 25 x 0.2ml tube block or a 16 x 0.5ml block. Heating and cooling are accomplished electronically by Peltier units. Uniformity across the block is ±0.4ºC at 55°C, ensuring that all samples are exposed to the same temperature conditions. Block heating and cooling are driven by an algorithm that accurately simulates sample temperature. The MultiGene II is supplied with a heated lid for oil-free cycling. For applications where the heated lid is not required, it can be turned off.
Cycling parameters are entered using the convenient numerical keypad. Programs can be run immediately upon entering, or up to 99 programs can be saved to the memory for later use. In addition to the standard parameters of time and temperature, the software also allows for successive time and temperature increments and decrements (for touchdown amplification and auto-extension), auto-restart after a power failure, end of cycling elongation steps and extended period soaks at 4ºC. The cycler can also be used for holding specific temperatures, allowing it to take the place of a traditional water bath, heat block or incubator.
