How to make Polyclonal Antibodies

Polyclonal antibodies (PA or pAbs ) are produced by different type of B cells lineages. Therefore, this type of antibodies has different structure even if they bind to the same antigen, just slightly different region (epitope).

Elisa Principle

Polyclonal Antibodies production

The production of the polyclonal antibodies takes just a few easy steps:
– Introducing the antigen (for example Occludin (OCLN)) to the organism that has functional Immune system. Those organisms are called hosts. Different type of hosts could be used such as Rabbit, Goat, Mouse, Sheep, guinea pigs, etc.
– Once the antigen is introduced into the host and antibodies formed (waiting period around 6 weeks) and a sample of blood is drawn. This sample contains polyclonal antibodies specific to the antigen Occludin (OCLN). This antigen could be species specific, there for the antibody would be Organism Species specific.
– Once isolated the polyclonal antibody undergoes to variety of purification technics such as Antigen-specific affinity chromatography followed by Protein A affinity chromatography, Protein G purified, etc.
If you would like to receive more information, regarding the methods that were applied for purification on a specific antibody, please do not hesitate to contact Gentaur experts. We guarantee that our products are produced in certified laboratories and manufacturing facilities.

Polyclonal antibodies Applications

We do understand, how important is to obtain high quality polyclonal antibody that had been validate for the specific application prior to investing time and efforts to implement it into your research. Therefor we offer high percentage of polyclonal antibodies validate for varieties of applications such as: Western Blot (WB), Immunofluorescence (IF), Native Chromatin Immunoprecipitation (ChIP), ELISA, Immunohistochemistry (IHC), Immunocytochemistry (ICC).For all the tested applications we could offer additional information on proposed dilution and optimal protocol for performing the analyses.

Storage conditions

Generally, the antibody would shipped at +4 °C (blue ice packs). Upon receving, freeze at -20 °C. For longer periods of time we recommend keeping the vial frozen at -40 °C or -80 °C. Avoid cycles of freezing and thawing as they may denaturate the polypeptide chains of the antibody. Due to transportation or handling reasons, small amounts of the antibody might get caught on the lid or walls of the vial. We recommend you to briefly cetrifuge the vial prior to use to gather the content on the bottom.
